Introduction
Identifying and isolating high-value antibody-secreting cells from mixed populations remains a major challenge in therapeutic discovery. Traditional screening methods often lack the precision and scalability needed to efficiently pinpoint the most productive cells. Cyto-Mine® Chroma overcomes these limitations by integrating multiplexed fluorescence detection with advanced picodroplet-based encapsulation.
In this study, we explore how Cyto-Mine® Chroma’s expanded laser and detection capabilities enable the precise screening and selection of specific antibody-secreting cells. Using a 4-color serial gating strategy combined with Förster Resonance Energy Transfer (FRET) assays, this approach enhances productivity screening, streamlines clone selection, and accelerates antibody discovery.

∙ Far Red Fluorescence (eFL670) – Identifies Antibody Secreting Cell “A”
∙ Orange Fluorescence (DL594), Green Fluorescence (DL488) – FRET pairs to identify IgG production from the antibody secreting cells.
ENCAPSULATION TECHNOLOGY enables functional screening of antibody secreting cells by creating millions of picolitre sized reaction vessels in a single experiment.
OFF-LINE FLUORESCENT IMAGING confirms the presence of only the sorted picodroplets that meets the selection criteria and has been dispensed from the Cyto-Mine® Chroma to a microplate.
Study Insights
By expanding fluorescence detection to four channels and introducing serial gating during sorting, Cyto-Mine® Chroma provides a more refined approach to screening and selecting target cells of interest. The FRET responses in conjunction with fluorescent labeling can be used to zero in on the picodroplets containing the specific antibody secreting cells within the mixed population.
Conclusion
Cyto-Mine® Chroma represents a major leap forward in single-cell analysis. This study showcases its ability to improve assay resolution, increase screening efficiency, and support next-generation therapeutic development.
